目的 :为了进一步观察鹿角方抗心肌纤维化的作用及其可能作用机制。方法 :建立腹主动脉狭窄所致充血性心力衰竭心肌肥厚大鼠模型 ,观察左室质量 (LVMI) ,采用RT -PCR方法观察心肌Ⅰ型和Ⅲ型胶原mRNA的表达 ,并采用放射免疫分析方法检测血浆和心肌血管紧张素Ⅱ (AngⅡ)水平。结果 :模型组LVMI明显高于假手术组 (P <0 0 0 1 ) ,鹿角方组LVMI与模型组比较有明显下降 (P <0 0 1 ) ;模型组Ⅰ型胶原mRNA的表达假手术组显著升高 (P <0 0 5) ,鹿角方组较模型组明显下降 (P <0 0 5) ;模型组Ⅲ型胶原mRNA与假手术组比较有显著升高 (P <0 0 5) ,鹿角方组较模型组显著下降 (P<0 0 5)。模型组血浆和左室心肌AngⅡ水平较假手术组显著升高 (P <0 0 0 1 ) ,鹿角方组明显降低血浆AngⅡ (P <0 0 1 )和左室心肌AngⅡ水平 (P <0 0 0 1 )。结论 :心肌纤维是腹主动脉狭窄所致充血性衰竭心肌肥厚的一个重要的病理改变 ,鹿角方具有逆转充血性心力衰竭心肌纤维化的作用。
Objective: To further observe the effect of Lujiao Fang on myocardial fibrosis and its possible mechanism. METHODS: A rat model of cardiac hypertrophic congestive heart failure caused by abdominal aortic stenosis was established. The left ventricular mass (LVMI) was observed. RT-PCR was used to observe the mRNA expression of type I and type III collagen in the myocardium. Radioimmunoassay was used. Plasma and myocardial angiotensin II (AngII) levels were measured. RESULTS: The LVMI in the model group was significantly higher than that in the sham operation group (P < 0.01). The LRMI in the Lujiaofang group was significantly decreased compared with the model group (P <0 01). The expression of type I collagen mRNA in the model group was sham-operated group. Significantly increased (P <0 05), Lujiao prescription group was significantly lower than the model group (P <0 05); Model group III collagen mRNA was significantly higher than the sham group (P <0 05). The deer-angle group significantly decreased compared with the model group (P<0 05). The plasma levels of Ang II in the plasma and left ventricular myocardium in the model group were significantly higher than those in the sham operation group (P <0 0 01). The Ang II group significantly decreased plasma Ang II (P <0 01) and left ventricular myocardial Ang II levels (P <0. 0 1 ). Conclusion: Myocardial fiber is an important pathological change of myocardial hypertrophy due to congestive heart failure caused by abdominal aortic stenosis. Lujiao Fang has the effect of reversing myocardial fibrosis in congestive heart failure.
