目的:探讨丹蒌片联合瑞舒伐他汀抗动脉粥样硬化的作用及机制。方法:雄性日本大耳白兔40只,随机分为正常对照组(CL组)、模型组(M组)、丹蒌片组(DL组)、瑞舒伐他汀组(RS组)、丹蒌片联合瑞舒伐他汀组(联合治疗组,IT组),每组8只。正常对照组饲喂普通饲料,余各组腹主动脉球囊损伤后高脂饲料喂养制备动脉粥样硬化模型;RS组[1 mg/(kg·d)]、DL组[0.5 g/(kg·d)]及IT组[0.5 g/(kg·d)]加[1 mg/(kg·d)]除高脂饲料喂养外喂饲相应药物。连续给药9周后处死,测各组血脂、一氧化氮(NO)和一氧化氮合酶(e NOS)的含量;HE染色观察主动脉病理学改变及内膜增生水平,Western blot检测腹主动脉中p-PI3K和p-Akt的蛋白表达水平。结果:M组中总胆固醇(TC)和低密度脂蛋白(LDL)度均显著高于CL组(均P<0.05);NO和e NOS显著降低(均P<0.05),病理学显示内膜增厚,斑块面积增加。Western blot显示腹主动脉组织中p-PI3K、p-Akt的蛋白表达水平均低于CL组(均P<0.05)。DL组、RS组和IT组与M组相比均降低TC、LDL,升高NO、e NOS水平,上调PI3K、p-Akt的蛋白表达(均P<0.05),IT组的效果最佳(均P<0.05)。结论:丹蒌片联合瑞舒伐他汀具有强化降低兔血脂、改善内皮细胞功能,抗动脉粥样硬化的作用,其作用机制可能与激活PI3/Akt信号通路相关。
Objective: To investigate the anti-atherosclerotic effect and mechanism of dancao tablet combined with rosuvastatin. Methods: Forty male Japanese white rabbits were randomly divided into normal control group (CL group), model group (M group), Danxuan tablet group (DL group), rosuvastatin group (RS group) Tablets combined with rosuvastatin group (combination therapy group, IT group), 8 in each group. The normal control group was fed with common diet, and the rest rabbits in each group were fed with high-fat diet after abdominal aorta balloon injury to prepare atherosclerosis model; RS group [1 mg / (kg · d)], DL group [0.5 g / · D)] and IT group [0.5 g / (kg · d)] plus [1 mg / (kg · d)] were given corresponding drugs except for high fat diet. After 9 weeks of continuous administration, the rats were sacrificed and the contents of blood lipid, nitric oxide (NO) and nitric oxide synthase (eNOS) in each group were measured. Pathological changes of aorta and intimal hyperplasia were observed by HE staining. The level of p-PI3K and p-Akt protein expression in the aorta. Results: The levels of total cholesterol (TC) and low density lipoprotein (LDL) in group M were significantly higher than those in group CL (all P <0.05), while NO and eNOS were significantly lower (all P <0.05) Thickening, plaque area increased. Western blot showed that the protein expression levels of p-PI3K and p-Akt in abdominal aorta were lower than those in CL group (all P <0.05). The levels of TC, LDL, NO and eNOS, the protein expressions of PI3K and p-Akt in DL group, RS group and IT group were significantly lower than those in M group (all P <0.05) All P <0.05). Conclusion: The combination of danji tablet and rosuvastatin has the effects of decreasing blood lipid, improving endothelial cell function and anti-atherosclerosis in rabbits. Its mechanism may be related to the activation of PI3 / Akt signaling pathway.
