目的探讨正常人眼视网膜色素上皮(RPE)细胞毒蕈碱1型(M1)受体的表达情况和M1受体对维持RPE细胞功能的作用及其与近视发生、发展的关系。方法采用已建立的RPE细胞的分离和培养方法建立细胞培养株。取传3~5代生长良好的细胞,应用逆转录聚合酶链反应(RT-PCR)、间接免疫荧光法(IIF)检测培养的RPE细胞中M1受体的表达情况。结果培养的RPE细胞呈多边形,平均分裂时间约2~3d,细胞内含有棕黄色素颗粒,随着细胞的分裂,细胞内的色素颗粒稀释和变少。RT-PCR和间接免疫荧光法检测,显示体外培养的人RPE细胞有M1受体表达。结论人RPE细胞有M1受体的分布。M1受体对维持RPE细胞的功能起重要作用。M1受体拮抗剂玻璃体腔注射以延缓近视的发生、发展机制可能与其抑制RPE细胞功能有关。
Objective To investigate the expression of muscarinic type 1 (M1) receptor in normal human retinal pigment epithelium (RPE) cells and the role of M1 receptor in maintaining RPE cell function and its relationship with the occurrence and development of myopia. Methods The cell culture strains were established by the methods of isolation and culture of established RPE cells. The well-grown cells were passaged from passage 3 to passage 5, and the expression of M1 receptor in cultured RPE cells was detected by reverse transcription-polymerase chain reaction (RT-PCR) and indirect immunofluorescence (IIF). Results The cultured RPE cells were polygons with an average time of 2-3 days. The cells contained yellow pigment granules. With the cell division, the pigment granules in the cells were diluted and decreased. RT-PCR and indirect immunofluorescence assay showed that human RPE cells cultured in vitro had M1 receptor expression. Conclusion Human RPE cells have M1 receptor distribution. The M1 receptor plays an important role in maintaining the function of RPE cells. M1 receptor antagonist intravitreal injection to delay the occurrence of myopia, the development mechanism may be related to its inhibition of RPE cell function.
