目的探讨Octreotide(OA)玻璃体腔注射对兔视网膜组织结构和功能的影响。方法健康的成年有色素兔24只24眼随机分为实验组(A)和对照组(B),实验组玻璃体腔注射OA,根据给药剂量不同分为A1(0.2mg)、A2(0.6mg)、A3(2.0mg)3组,对照组玻璃体腔注射复方氯化钠注射液,注药后1、2、4周分别对所有实验对象进行眼压、裂隙灯、眼底及ERG检查,4周后摘出眼球,光学显微镜下观察视网膜的组织形态结构。结果 2周后A3组2只动物出现晶状体混浊,A3组光镜下可见视网膜结构改变。实验组眼压有一过性升高,但与对照组相比无统计学差异。实验组3组ERGb波振幅均有降低,与对照组相比有统计学差异,下降幅度随剂量增加而增加,2周时下降幅度最大,到观察末期A1、A2组有部分回升,而A3组变化不大。结论兔眼玻璃体腔注射OA剂量<0.6mg时是相对安全的。
Objective To investigate the effect of Octreotide (OA) intravitreal injection on the structure and function of rabbit retina. Methods Twenty-four eyes of healthy adult pigmented rabbits were randomly divided into experimental group (A) and control group (B). The experimental group received intravitreal injection of OA and were divided into A1 (0.2mg), A2 (0.6mg ) And A3 (2.0mg). The control group was injected intravitreally with compound sodium chloride injection. The intraocular pressure, slit lamp, fundus and ERG examination were performed on all the experimental subjects at 1, 2 and 4 weeks after injection. After the removal of the eye, observe the morphological structure of the retina under a light microscope. Results After 2 weeks, two animals in group A3 appeared opacification of the lens, and the retinal structure was observed under light microscope in group A3. The experimental group intraocular pressure had a transient increase, but no significant difference compared with the control group. The amplitudes of ERGb wave in experimental group 3 were decreased, compared with the control group, the amplitude of decrease was increased with the increase of dose, and the amplitude of ERGb wave decreased the most in the second week. Has not changed much. Conclusions Intraocular injection of OA in rabbits eye is relatively safe when the dosage is less than 0.6mg.
