目的 探讨胃液对膀胱癌生长抑制、细胞凋亡及bcl 2 /bax表达的影响。方法 采用细胞培养、四唑盐比色试验 (MTT)、DNA提取及电泳、流式细胞仪和免疫组织化学等方法 ,观察胃液对膀胱癌细胞系生长抑制、细胞凋亡及bcl 2 /bax表达的影响。结果 高浓度胃液 (19~ 3 8mmol/L)抑制膀胱癌细胞系的生长繁殖 ,低浓度胃液 (0 .5 94~ 9.5 0 0mmol/L)则促进膀胱癌细胞系的生长繁殖。胃液能诱导体外培养的膀胱癌细胞凋亡。膀胱癌细胞系BIU 87中bcl 2、bax均有表达 (分别为 63 .5 %和 41.4% ) ,经胃液、稀盐酸或阿霉素处理后 ,bcl 2表达下降 (分别为 3 4 .8%、3 7.3 %和 2 9.6% ) ,bax表达上升 (分别为 49.3 %、47.1%和 5 2 .2 % ) ,bcl 2 /bax比值小于 1。结论 胃液能抑制膀胱癌的生长 ,诱导其凋亡
Objective To investigate the effect of gastric juice on the growth inhibition, apoptosis and the expression of bcl 2 / bax in bladder cancer. Methods The growth inhibition, apoptosis and bcl 2 / bax expression of bladder cancer cell lines were observed by cell culture, MTT assay, DNA extraction and electrophoresis, flow cytometry and immunohistochemistry. Impact. Results High concentrations of gastric juice (19 ~ 38mmol / L) inhibited the growth and proliferation of bladder cancer cell lines. Low concentrations of gastric juice (0.594 ~ 9.5 0 0mmol / L) promoted the growth and proliferation of bladder cancer cell lines. Gastric juice can induce apoptosis of bladder cancer cells cultured in vitro. The expressions of bcl 2 and bax in bladder cancer cell line BIU 87 were 63.5% and 41.4%, respectively. After treatment with gastric juice, dilute hydrochloric acid or doxorubicin, the expression of bcl 2 decreased (34.8% , 33.3% and 29.6% respectively), bax expression increased (49.3%, 47.1% and 52.2% respectively), and the ratio of bcl2 / bax was less than 1. Conclusion Gastric juice can inhibit the growth of bladder cancer and induce its apoptosis
