甘蔗植物组织内的多酚类、色素类等物质是干扰RAPD扩增反应的主要成分。本研究以CTAB法和SDS法为基础,着重对抗氧化剂(PVP等)、酶(蛋白酶K、RNaseA)和酚抽提等因素对提取DNA的质量及其RAPD的影响进行了比较筛选。结果表明:①甘蔗DNA提取质量和纯度与RNaseA、抗氧化剂等处理直接相关,蛋白酶K、CTAB和SDS的处理无明显差异;②对于甘蔗RAPD反应,模板DNA中含有一定量的RNA和蛋白质,其扩增产物无明显差异,但模板DNA的降解程度和含有酚类、色素等是干扰甘蔗RAPD反应的主要因子。进一步分析讨论了甘蔗RAPD扩增的有效的DNA提纯方法和影响因子。
Polyphenols, pigments and other substances in sugarcane plant tissues are the main components that interfere with the RAPD amplification reaction. Based on the CTAB method and the SDS method, we compared the effects of antioxidants (PVP, etc.), enzymes (Proteinase K, RNase A) and phenol extraction on the quality of extracted DNA and its RAPD.
