目的:探讨左归丸防治骨质疏松症的机理。方法:以卵巢切除所致的骨质疏松大鼠为模型,采用骨组织形态计量学方法测定胫骨骨小梁体积百分比(TBV%)、骨小梁吸收表面百分比(TRS%)和骨小梁形成表面百分比(TFS%),采用放射免疫分析法测定外周血清中雌二醇(E_2)含量,分别采用小鼠胸腺细胞检测法和依赖株细胞增殖反应法测定白细胞介素-1(IL-1)和白细胞介素-6(IL-6)的活性。结果:左归丸能使胫骨TBV%显著增高,使TRS%和TFS%显著降低。大鼠切除卵巢后,E_2含量大幅度降低,而IL-1和IL—6活性显著增高。左归丸对E_2含量无显著性影响,但对IL—1和IL—6活性有明显的抑制作用。结论:左归丸能够抑制IL-1和IL-6的活性是其防治骨质疏松症的机理之一。
Objective: To explore the mechanism of Zuogui Pills in preventing and treating osteoporosis. METHODS: The osteoporotic rat model induced by ovariectomy was used to determine the percentage of trabecular bone volume (TBV%), percentage of trabecular bone absorptive surface (TRS%) and trabecular bone formation using bone histomorphometry. Percentage of surface (TFS%), radioimmunoassay was used to determine the content of estradiol (E_2) in peripheral blood. IL-1 was measured by mouse thymocyte assay and cell proliferation assay. And interleukin-6 (IL-6) activity. RESULTS: Zuogui Pill can significantly increase the TBV% of tibia and significantly reduce TRS% and TFS%. After ovariectomy in rats, the content of E2 decreased significantly, while the activity of IL-1 and IL-6 increased significantly. Zuogui Pill had no significant effect on the content of E2, but it had a significant inhibitory effect on the activity of IL-1 and IL-6. Conclusion: Zuogui Pill can inhibit the activity of IL-1 and IL-6 is one of the mechanisms of its prevention and treatment of osteoporosis.
