目的探讨益气活血汤对慢性肾功能衰竭(CRF)大鼠结缔组织生长因子(CTGF)、血小板源性生长因子-B(PDGF-B)的影响。方法采用腺嘌呤制作大鼠CRF模型,将其分为正常对照组、模型对照组、科索亚组及模型中药高、中、低剂量组,治疗8周后,测量各组血肌酐(SCr)、尿素氮(BUN)、甘油三脂(TG)、胆固醇(CHO)、血清白蛋白(ALB)、血红蛋白(HB)水平,HE染色观察大鼠肾脏组织病理学变化,免疫组织化学SP法检测CTGF、PDGF-B在大鼠肾组织的表达。结果模型中药高、中、低剂量组及科索亚组SCr、BUN、CHO水平均低于模型对照组,ALB、HB高于模型对照组(P<0.05)。病理结果显示,各用药组大鼠的肾组织病理损伤明显轻于模型对照组,模型中药高剂量组损伤最轻。免疫组化结果显示,各用药组CTGF、PDGF-B在肾组织的表达均明显低于模型对照组(P<0.05)。结论益气活血汤可有效降低CRF大鼠SCr、BUN、CHO水平,升高ALB、HB水平,具有减轻大鼠肾组织的损伤、减少CTGF、PDGF-B在肾组织的表达、延缓CRF进展的作用。
Objective To investigate the effects of Yiqi Huoxue Decoction on connective tissue growth factor (CTGF) and platelet-derived growth factor-B (PDGF-B) in rats with chronic renal failure (CRF). Methods The rat CRF model was made using adenine. The rats were divided into normal control group, model control group, Kosoa group and model high, medium and low dose groups. After 8 weeks of treatment, serum creatinine (SCr) was measured. Levels of BUN, TG, CHO, ALB, and HB were observed by HE staining, and histopathological changes in kidneys were detected by immunohistochemical SP method. Expression of PDGF-B in rat kidney tissue. Results The levels of SCr, BUN, and CHO in the high, medium, and low dose groups and Corso Group were lower than those in the model control group. ALB and HB were higher than the model control group (P<0.05). The pathological results showed that the pathological damage of the kidneys of rats in each drug group was significantly lighter than that of the model control group, and the damage of the high dose group was the lightest. The results of immunohistochemistry showed that the expression of CTGF and PDGF-B in the renal tissue of each drug group was significantly lower than that of the model control group (P<0.05). Conclusion Yiqihuoxue decoction can effectively reduce the levels of SCr, BUN and CHO in CRF rats and increase the levels of ALB and HB. It can reduce the damage of renal tissue, decrease the expression of CTGF and PDGF-B in renal tissues, and delay the progression of CRF. effect.
