目的建立竹节参及其近缘人参属植物种间快速鉴别体系。方法采用RAPD技术获得DNA指纹图谱,根据图谱中特异性条带,绘制人工绘制品种鉴定图(MCID)。结果从16条随机引物中筛选出能稳定扩增并且具有特异性条带的4条引物构建DNA指纹图谱,共扩增出46条带,其中多态性条带42条,多态性比率为91.30%。利用其中的多态性条带,绘制出图谱关系分析图,构成竹节参及其近缘植物的人工绘制品种鉴定图。结论该方法快速简单,结果直观可靠,为竹节参及近缘品种的初步鉴别提供了一种有效可行的方法。
OBJECTIVE To establish a rapid differential identification system between species of Panax ginseng and its related ginseng. Methods DNA fingerprinting was obtained by RAPD technique. According to the specific bands in the map, artificial identification map (MCID) was drawn. Results Four primers were screened out from 16 random primers to construct a DNA fingerprinting. A total of 46 bands were amplified, of which 42 were polymorphic. The polymorphism rate was 91.30%. The polymorphism bands were used to draw the map of the relationship between the maps and constitute the artificial drawing variety identification map of the bamboo reference plant and its related plants. Conclusion The method is rapid and simple, the results are intuitive and reliable, which provides an effective and feasible method for the initial identification of bamboo reference and related species.
