用巨尾按试管培养的节段作为种质,用海藻酸钠包裹成胶丸,埋放琼脂培养基内,在一般培养室条件下保存10个月时,再生率可达52%。找出了最适宜的保存条件为1.5%~2.0%海藻酸钠,0.5%蔗糖和1/4浓度的MS基本培养基和大量无机元素;光照是必要的外部条件;蔗糖是制约保存种质活力和再生产的关键因子。研究还发现保存的种质胶丸颜色由绿色转为白色或黑色是种质丧失活力的两大主要过程。进一步研究了种质保存中与代谢有关的几个生理参数:呼吸、叶绿素和可溶蛋白含量以及多酚氧化酶的变化,为常温下以胶丸形式、在微生长状态下保存种质提供了生理、生化依据。
