目的 探讨周期蛋白A(cyclinA)在原发性肝癌 (HCC)中的表达、临床意义及其与乙型肝炎病毒x基因整合之间的关系。方法 分别采用PCR、RT PCR、Westernblot检测HCC及其相邻非肿瘤组织中的cyclinA基因、mRNA和蛋白的表达 ;采用PCR和Southernblot检测HCC标本中HBx基因的整合。结果 35例HCC标本中 ,cyclinA基因扩增率为 2 .8% (1/ 35 ) ,mRNA过表达率为 4 5 .7%(16 / 35 ) ,蛋白过表达率为 6 0 .0 % (2 1/ 35 )。cyclinA蛋白过表达与患者年龄、肿瘤大小以及HBx基因整合之间存在统计学联系。结论 cyclinA的表达失控在肝细胞癌变的早期发生 ,可能是乙型肝炎病毒 (HBV)影响肝细胞正常细胞周期的重要途径之一。
Objective To investigate the expression of cyclin A in primary hepatocellular carcinoma (HCC) and its clinical significance and its relationship with hepatitis B virus x gene integration. Methods The expression of cyclinA mRNA and protein in HCC and its adjacent non-tumor tissues were detected by PCR, RT PCR and Western blot, respectively. The integration of HBx gene in HCC specimens was detected by PCR and Southern blot. Results Among 35 HCC specimens, the rate of cyclin A gene amplification was 2.8% (1/35), the overexpression rate of mRNA was 45.7% (16/35) and the overexpression rate of protein was 60.0% 2 1/35). There was a statistically significant correlation between cyclinA protein overexpression and patient age, tumor size, and HBx gene integration. Conclusion The out-of-control of cyclinA expression may be one of the important pathways of Hepatitis B virus (HBV) affecting the normal cell cycle of hepatocytes.
