目的探讨山东地区汉族人群CD14基因rs2569190和rs2563298多态性与类风湿关节炎(RA)易感性的关联性。方法采用高分辨率熔解曲线方法对256例RA患者及331例健康对照者进行基因分型,计算各位点基因型及等位基因频率,检测血沉(ESR)、C反应蛋白(CRP)、抗环瓜氨酸多肽抗体(CCP)、葡萄糖6磷酸异构酶(GPI)等指标,比较两组各位点之间基因型及等位基因频率分布差异。结果 CD14基因rs2569190等位基因频率在RA组和对照组间差异有统计学意义(P=0.017),基因型分布在两组间差异无统计学意义(P=0.058);rs2563298基因型及等位基因频率在两组间差异无统计学意义(P=0.444,0.599)。经Bonferroni多重检验校正后,rs2569190等位基因频率在RA组与对照组间差异有统计学意义(校正后P=0.034)。CD14基因rs2569190和rs2563298在年龄、性别、病程、DAS28及各项指标上不同基因型间差异无统计学意义(P>0.05)。结论在山东地区汉族人群中CD14基因rs2569190多态性与类风湿关节炎易感性可能有关联。
Objective To investigate the association between rs2569190 and rs2563298 polymorphisms of CD14 gene and susceptibility to rheumatoid arthritis (RA) in Shandong Han population. Methods The genotypes of 256 patients with RA and 331 healthy controls were genotyped by high resolution melting curve method. The frequencies of genotypes and alleles at each site were calculated. ESR, CRP, Citrulline peptide antibody (CCP), glucose 6 phosphate isomerase (GPI) and other indicators were compared between the two groups of genotypes and allele frequency distribution differences. Results The frequency of rs2569190 allele of CD14 gene was significantly different between RA group and control group (P = 0.017). There was no significant difference in genotype distribution between the two groups (P = 0.058). The rs2563298 genotype and allele There was no significant difference in gene frequency between the two groups (P = 0.444, 0.599). After Bonferroni multiple test calibration, rs2569190 allele frequency was significantly different between RA group and control group (P = 0.034 after adjustment). The genotypes of CD14 rs2569190 and rs2563298 in age, sex, duration of disease, DAS28 and other indicators had no significant difference (P> 0.05). Conclusion The rs2569190 polymorphism of CD14 gene in Shandong Han population may be associated with the susceptibility to rheumatoid arthritis.
