目的:优化DM301大孔吸附树脂纯化补肾方总黄酮的工艺条件及参数。方法:以总黄酮和指标成分含量(二苯乙烯苷、淫羊藿苷和柚皮苷)为指标,采用正交试验方法考察上样量、径高比、洗脱溶剂用量和洗脱速度4个因素对纯化效果的影响,并进行了优化工艺的重复性试验和纯化前后指标成分含量比例变化的考察。结果:经优化的纯化工艺条件为:树脂药材质量比为2∶1,树脂柱径高比为1∶10,洗脱溶剂量为4BV,洗脱速度为2.3BV.h-1,经纯化后总黄酮纯度达50%以上,总黄酮的保留率为76.30%,纯化前后3个指标成分的含量比例基本相同。结论:经该工艺纯化后明显提高了总黄酮的纯度,减少了纯化固形物的量,从而减少服用量。
Objective: To optimize the process conditions and parameters for the purification of total flavonoids from Bushen Recipe by DM301 macroporous adsorption resin. Methods: The content of total flavonoids and indicator components (stilbene glycoside, icariin and naringin) were used as indexes to investigate the effects of sample volume, diameter-height ratio, elution solvent amount and elution speed 4 The factors affecting the purification effect were investigated. The repeatability of the optimization process and the ratio of the content of the indicator components before and after purification were investigated. Results: The optimized purification conditions were as follows: the mass ratio of resin to drug was 2:1, the diameter ratio of resin was 1:10, the amount of elution solvent was 4BV and the elution rate was 2.3BV.h-1. After purification The purity of total flavonoids reached more than 50%, the retention rate of total flavonoids was 76.30%. The contents of three index components before and after purification were basically the same. Conclusion: The purity of total flavonoids is obviously increased after purification by the process, the amount of purified solid is reduced, and the dosage is reduced.
