[摘要] 目的 探讨DyrklB在宫颈癌组织中的表达及其临床意义。 方法 利用免疫组化检测DyrklB在60例宫颈癌组织、60例宫颈癌癌旁组织和60例正常宫颈组织中的表达,分析DyrklB在宫颈癌组织中的表达状况以及与宫颈癌临床参数的关系。 结果 Dyrk1B 在宫颈癌组织中的表达(60/60,100%)明显高于癌旁组织(26/40,65%)和正常宫颈组织(16/40,40%),三组间比较差异有统计学意义(χ2=88.21,P<0.01)。Dyrk1B的强阳性率与宫颈癌的组织学分化程度有明显相关性(P<0.05),与宫颈癌患者的年龄、临床分期和有无淋巴转移等无明显相关性(P>0.05)。 结论 DyrklB在宫颈癌中高表达,提示其可能参与了肿瘤的发生和发展,并有望成为宫颈癌早期诊断的标志物和治疗的靶基因。

Expression and clinical significance of Dyrk1B in cervical carcinoma

[Abstract] Objective To investigate the expression of DyrklB in cervical cancer and its clinical significance. Methods Immunohistochemical methods were used to detect the expression levels of DryklB in 60 cases cervical carcinoma tissues,60 cases of cervical cancer adjacent tissues and 60 cases normal cervical tissues. DryklB expression in cervical carcinoma and relationship with clinical parameters of cervical carcinoma. Results The expression of Dyrk1B in cervical cancer tissues was significantly higher than that in cervical cancer adjacent tissues and normal cervical tissues,and there was statistical significance between the three groups(χ2=88.21,P<0.01). There was a significant correlation between the strong positive rate of Dyrklb and degree of histological differentiation(P<0.05),but there was no correlation between the strong positive rate of Dyrklb and patient’s age,clinical stage and lymph node metastasis of cervical carcinoma(P>0.05). Conclusion The expression level of Dyrk1b in cervical carcinoma tissues is high,which indicates that it may participate in the occurrence and development of cervical carcinoma ,and it may become a tumor marker for clinical early diagnosis and a new target gene for treatment of cervical carcinoma.

双重特异性酪氨酸磷酸化调节激酶1B(dual-specificity tyrosine-phosphorylation regulated kinase 1B,Dyrk1B;又称Mirk)属Minibrain Dyrk家族成员,是一种苏氨酸/丝氨酸蛋白激酶,该蛋白基因定位在人类19号染色体q12-13.11上,其功能是特异性磷酸化下游的双链酪氨酸[1],是一种特异性的双链酪氨酸磷酸化激酶[2],最初是从结肠癌细胞株[3]和人类睾丸组织[4]中分离出来的。DyrklB可在翻译过程中完成自身磷酸化,并同时有磷酸化其他丝氨酸、苏氨酸的能力[5],研究证实,Dyrk1B在细胞转录、细胞周期及细胞凋亡的调节中发挥着重要的作用。通过血清饥饿诱导静止状态,同时通过siRNA等方法沉默Dyrk1B的表达,可减少多种肿瘤细胞系的集落形成,并增强化疗药物诱导的细胞凋亡。Dyrk1B 除在人类的脑组织及肌肉组织中有表达外,在大多数正常组织中呈低表达或不表达[6],但在某些恶性肿瘤组织中呈高水平表达,如胰腺癌[7,8]、结肠癌[9]、肺癌[10,11]、卵巢癌[12-14]、横纹肌肉瘤[15]、前列腺癌[16]等。Dyrk1B在宫颈癌细胞中的表达水平未见报道,本研究旨在应用免疫组化方法检测宫颈癌组织、癌旁组织及正常宫颈组织中Dyrk1B的表达情况,探讨Dyrk1B与宫颈癌发生、发展、分化程度、侵袭的相关性及其临床意义,现报道如下。
