目的:观察芍药甘草汤有效组分(SGM)对慢性压迫性损伤(CCI)大鼠机械痛敏和热痛敏的影响,并研究SGM的给药时机和是否有超前镇痛作用。方法:所有大鼠用机械缩足反射阈值(MWT)和热缩足潜伏期(TWL)来分别评价大鼠机械痛敏和热痛敏。SD大鼠随机分为9组,即假手术组、CCI组和空白组,BN组和BS组,于术前7天开始灌胃给予0.9%生理盐水或SGM120mg/kg,每天2次,直至手术当天结束;AN组、AS组为后给药组,于术后第1天给予直至术后第7天;BAN组和BAS组术前7天前开始直至术后第7天。各组分别在术前、术后1、3、7天分别测定MWT和TWL。结果:和空白组和假手术组相比,CCI组术后MWT及TWL明显下降(P<0.01)。BS、AS和BAS组与各生理盐水组相比,MWT和TWL在术后1、3和7天均明显增加(P<0.01)。BS组和AS组相比,术后第1天的MWT和TWL均明显增加(P<0.01),但到术后第7天,AS组的MWT和TWL均高于BS组(P<0.01),BAS组和AS组相比,术后第1天的MWT和TWL均明显增加(P<0.01),但到术后第7天,两者无明显差异。结论:CCI大鼠从术后1天形成稳定的机械痛敏和热痛敏,SGM能抑制CCI大鼠的机械痛敏和热痛敏作用,术前给予SGM对NP有超前镇痛作用,镇痛作用并能持续数天。和术后用药相比,术前后给药组镇痛作用出现更快。
OBJECTIVE: To observe the effect of SGM on mechanical pain and thermal hyperalgesia in rats with chronic constriction injury (CCI), and to investigate the timing of SGM administration and the preemptive analgesic effect. Methods: All rats were subjected to mechanical hyperalgesia and thermal hyperalgesia by mechanical contractive threshold (MWT) and thermal contraction foot latency (TWL), respectively. Compared with AS group, MWT and TWL were significantly increased on the first postoperative day (P <0.01), but the MWT and TWL of the AS group were significantly higher than those of the BS group on the seventh postoperative day (P <0.01) Compared with AS group, the MWT and TWL on the first postoperative day were significantly increased (P <0.01), but there was no significant difference between the BAS group and the AS group on the seventh postoperative day. CONCLUSIONS: CCI rats form stable mechanical and thermal hyperalgesia one day after operation. SGM can inhibit the mechanical and thermal hyperalgesia effects of CCI rats. Preoperative administration of SGM has a preemptive analgesic effect on NP and analgesic effect And can last for days. Compared with postoperative medication, the analgesic effect appeared faster in the preoperative and postoperative groups.
