目的:进一步探讨糖皮质激素防治肾间质纤维化的机理。方法:在成功分离培养人肾间质成纤维细胞基础上,通过流式细胞仪分析、dUTP标记DNA断裂点显色及电镜技术等方法,观察甲基强的松龙对肾间质成纤维细胞编程性死亡的影响。结果:甲基强的松龙处理组编程性死亡的细胞百分数明显高于对照组,且呈时间依赖性。结论:甲基强的松龙可促进人肾间质成纤维细胞的凋亡,可能是糖皮质激素防治肾间质纤维化的重要机理之一
Objective: To further explore the mechanism of glucocorticoid in prevention and treatment of renal interstitial fibrosis. Methods: Based on the successful isolation and culture of human renal interstitial fibroblasts, the effects of methylprednisolone on renal interstitial fibroblasts were observed by flow cytometry, dUTP-labeled DNA breakpoint colorimetry and electron microscopy. Impact of programmed death. Results: The percentage of apoptotic cells in methylprednisolone treatment group was significantly higher than that in control group and in a time - dependent manner. CONCLUSION: Methylprednisolone can promote the apoptosis of human renal interstitial fibroblasts and may be one of the important mechanisms of glucocorticoid in prevention and treatment of renal interstitial fibrosis
