目的观察三氧化二砷(As2O3)、干扰素(IFN-α1、IFN-α2、IFN-β)及其两者联合对人类骨髓瘤细胞株(RPMI8226)的增殖的影响。方法以四甲基偶氮唑盐微量酶反应比色法(MTT)测定RPMI8226细胞的增殖活性,并观察不同类型、不同浓度干扰素和亚砷酸对RPMI8226细胞增殖的影响以及两者联合对RPMI8226细胞的增殖作用。结果As2O3对RPMI8226细胞增殖有显著的抑制作用;IFN-α1对RPMI8226细胞生长无抑制作用,当IFN-α2浓度大于500 kU/L和IFN-β浓度大于1000 kU/L时对RPMI8226细胞增殖有抑制作用(P<0.05);且IFN-α2与As2O3联合对RPMI8226细胞增殖抑制有协同作用(P<0.05)。结论As2O3对骨髓瘤细胞增殖有抑制作用,而干扰素须达到一定浓度才对RPMI8226细胞增殖有抑制作用。
Objective To observe the effects of combination of As2O3 and IFN-α1, IFN-α2 and IFN-β on the proliferation of human myeloma cell line RPMI8226. Methods The proliferative activity of RPMI8226 cells was determined by MTT assay. The effects of different types of interferon and arsenious acid on the proliferation of RPMI8226 cells were observed. Cell proliferation. Results As2O3 had a significant inhibitory effect on the proliferation of RPMI8226 cells. IFN-α1 had no effect on the growth of RPMI8226 cells. When the concentration of IFN-α2 was more than 500 kU / L and the concentration of IFN-β was more than 1000 kU / L, the proliferation of RPMI8226 cells was inhibited (P <0.05). The combination of IFN-α2 and As2O3 had synergistic effect on the proliferation inhibition of RPMI8226 cells (P <0.05). Conclusion As2O3 can inhibit the proliferation of myeloma cells. However, when the concentration of interferon reaches a certain concentration, it can inhibit the proliferation of RPMI8226 cells.
