建立多角度激光光散射仪和体积排阻色谱法联用测定透明质酸相对分子质量及其分布的方法。考察MALLS-SEC法测定HA Mr的准确性和精密度,以及不同HA溶液浓度对测定的影响。结果表明:MALLS-SEC法的准确性好、精密度高,相对标准偏差小于2%;测定HA Mr时,采用0.1mg/mL的进样浓度较合适。操作简单、快速,结果准确,可用于HA的质量控制和应用研究。
To establish a method for the determination of molecular weight and distribution of hyaluronic acid by multi-angle laser light scattering and size exclusion chromatography. The MALLS-SEC method was used to determine the accuracy and precision of HA Mr and the effect of different concentrations of HA solution on the determination. The results showed that the accuracy of MALLS-SEC method was high, the precision was high, and the relative standard deviation was less than 2%. When the concentration of HA was about 0.1mg / mL, the injection concentration was more suitable. Simple, fast, accurate and can be used for quality control and applied research of HA.
