目的:研究蒙药库页悬钩子中总皂苷和齐墩果酸的含量测定方法及最佳采收期。方法:选用齐墩果酸为对照品,采用分光光度法,5%香草醛-冰醋酸和高氯酸显色,在波长547 nm处测定吸光度,计算不同采收期库页悬钩子中总皂苷含量;采用高效液相色谱法对齐墩果酸含量进行测定,Phenomsil C18色谱柱(4.6 mm×250 mm,5μm),柱温为室温(25℃),流动相甲醇-0.3%磷酸水溶液(80∶20),流速1 mL·min-1,检测波长220 nm。结果:不同采收期库页悬钩子中总皂苷和齐墩果酸含量发生明显变化。总皂苷在6~9月含量逐渐升高,9月含量最高,10月逐渐下降,齐墩果酸含量从8月开始逐渐升高,9月达最大值,10月下降。结论:该实验方法简便、快速、重复性好,可用于库页悬钩子总皂苷和齐墩果酸的含量测定,库页悬钩子宜于9月采集。
OBJECTIVE: To study the determination of total saponins and oleanolic acid in Rubus albopictus L. and the optimal harvest time. Methods: Oleanolic acid was used as the reference substance. The absorbance was measured at 547 nm by spectrophotometry, 5% vanillin-acetic acid and perchloric acid, and the total saponins The content of oleanolic acid was determined by high performance liquid chromatography (HPLC) on a Phenomsil C18 column (4.6 mm × 250 mm, 5 μm) with a mobile phase of methanol-0.3% phosphoric acid 20), flow rate 1 mL · min-1, detection wavelength 220 nm. Results: The contents of total saponin and oleanolic acid in Rubus macrospora were significantly changed in different harvest periods. The contents of total saponins gradually increased from June to September, with the highest content in September and the gradual decrease in October. The content of oleanolic acid increased gradually from August, reaching the maximum in September and declining in October. Conclusion: The method is simple, rapid and reproducible. It can be used for the determination of total saponins and oleanolic acid in raspberry leaves. The raspberry leaves should be collected in September.
