细胞凋亡在神经系统发育、神经系统疾病和外伤中扮演着重要角色。死亡受体不仅能触发细胞凋亡,还能促进细胞的生存和生长。最近研究显示,部分死亡受体在神经发育或退化等方面发挥着重要作用。死亡受体在帕金森病中具有神经保护的作用,在肌萎缩性脊髓侧索硬化和脑缺血性疾病中诱发凋亡前体的产生。这种不同的功能反映出在神经元和神经胶质细胞中死亡受体在转录和翻译信号通路下游的不同机制。本文就死亡受体在神经系统发育和疾病中的作用及其细胞内信号通路作一综述。
Apoptosis plays an important role in the development of nervous system, nervous system diseases and trauma. Death receptors not only trigger apoptosis, but also promote cell survival and growth. Recent studies have shown that some death receptors play an important role in neurodevelopment or degeneration. Death receptors have neuroprotective effects in Parkinson’s disease and induce the production of apoptotic precursors in amyotrophic lateral sclerosis and cerebral ischemic disease. This different function reflects a different mechanism of death receptors downstream of the transcriptional and translational signaling pathways in neurons and glial cells. This article reviews the role of death receptors in the development and progression of the nervous system and their intracellular signaling pathways.
