目的:观察天麻素对豚鼠噪声性声损伤预防作用及对耳蜗毛细胞SDH影响,同时探讨其可能的作用机理。方法:将实验用雄性红目纯白豚鼠30只,随机编3组,正常组、预防实验组(I_预)、预防对照组(I_对)。将除正常组外2组动物于隔音室内暴露于105dB SPL稳态白噪声下,暴露时间持续每天4小时,连续7天。预防实验组于噪声前3天开始给天麻素100mg/kg,腹腔注射,持续至噪声结束当天,预防对照组同期给予注射用水;测ABR以观察听阈值变化;光镜观察毛细胞琥珀酸脱氢酶(SDH)活性变化。结果:预防实验组ABR听阈值明显低于预防对照组(P<0.05);SDH染色细胞预防实验组明显优于预防对照组。结论:进一步证实天麻素能增加耳蜗毛细胞SDH活性,保护纹血管内皮细胞系统功能。对噪声性声损伤有一定的预防作用。
OBJECTIVE: To observe the preventive effect of gastrodin on noise-induced acoustic injury in guinea pigs and its effect on the SDH of cochlear hair cells, and to explore its possible mechanism. Methods: Thirty male Red Guinea pigs were randomly divided into 3 groups: normal group, prophylactic experimental group (I_pre) and prophylactic control group (I_). Except for the normal group, two groups of animals were exposed to 105 dB SPL steady-state white noise in the soundproof room, and the exposure time continued for 4 hours a day for 7 consecutive days. The prevention group began to inject gastrodin 100mg / kg three days before the noise into the abdominal cavity and continued to the end of the noise, and the control group was given water for injection at the same time. The ABR was used to observe the change of hearing threshold. Changes in enzyme (SDH) activity. Results: The threshold of ABR in experimental group was significantly lower than that of control group (P <0.05). SDH-stained cells prevention group was significantly better than that of control group. Conclusion: Gastrodin can further increase the SDH activity of cochlear hair cells and protect the function of the vascular endothelial cells. The noise of acoustic damage have a preventive effect.
