目的:研究5-氟尿嘧啶植入剂(5-Fuimplant5Ful)犬皮下植入的药代动力学。方法:用高效液相色谱法(HPLC)测定血浆5-Fu浓度。结果:犬皮下植入5-FuI20mg/kg后血药浓度—时间变化过程符合具有缓释的一房室模型,缓释半衰期T0.5r=52.7±10.9h,在体内维持有效浓度(≥1ug/mL)时间长达216~240h。结论:5-FuI皮下植入确具缓释作用和维持有效浓度时间长的优点。
Objective: To study the pharmacokinetics of subcutaneous implantation of 5-fluorouracil implant (5-Fuimplant5Ful) in dogs. Methods: Plasma 5-Fu concentration was determined by high performance liquid chromatography (HPLC). Results: The time-course of plasma concentration-time change after instillation of 5-FuI at 20mg / kg in canines was in accordance with the one-compartment model with sustained release. The half-life of half-life T0.5r = 52.7 ± 10.9h and the effective concentration in vivo (≥1ug / mL) as long as 216 ~ 240h. Conclusion: The subcutaneous implantation of 5-FuI has the advantage of sustained release and long-term maintenance of effective concentration.
