目的 探讨WH5 0 5 (丹参和山楂的提取物 )对动物体内血栓形成的影响和作用机制。方法 用大鼠颈总动脉—颈外静脉血流旁路和小鼠静脉注射胶原蛋白一肾上腺素造成血栓形成模型 ;采用流式细胞术考察WH5 0 5对人血小板经凝血酶诱导的P—选择素表达的影响。结果 WH5 0 5可明显抑制大鼠颈总动脉—颈外静脉血流旁路血栓形成 ,抑制率为 15 %~ 41% ;使胶原蛋白—肾上腺素诱导小鼠瘫痪的恢复率显著提高 ,恢复率为 6 4.3%~ 84.6 % ,阳性对照组为 80 %。人血小板经凝血酶刺激后 ,血小板表面表达P -选择素明显增加 ,用不同浓度WH5 0 5与血小板预孵 ,则可剂量依赖性地抑制血小板P -选择素的表达。结论 WH5 0 5对血小板性血栓形成具有抑制作用 ,该作用可能与抑制血小板P -选择素表达有关。
Objective To investigate the effect of WH5 0 5 (an extract of Salvia miltiorrhiza and Radix Salviae Miltiorrhizae) on thrombosis in animals and its mechanism of action. Methods Thrombus formation was induced by bypass of the common carotid artery and external jugular vein in rats and collagen-epinephrine injection in mice. Flow cytometry was used to investigate the P-selection induced by thrombin in human platelets by WH50.5. Effect of prime expression. Results WH5 05 could significantly inhibit the blood flow bypass thrombosis of common carotid artery and external jugular vein in rats, the inhibition rate was 15% to 41%. The recovery rate of collagen-adrenergic induced mice sputum was significantly increased, and the recovery rate was improved. It was 6 4.3% to 84.6% and the positive control group was 80%. When platelets were stimulated by human thrombin, platelet surface expression of P-selectin was significantly increased. Different concentrations of WH5 05 and platelet pre-incubation inhibited platelet P-selectin expression in a dose-dependent manner. Conclusion WH5 05 has an inhibitory effect on platelet thrombosis, which may be related to the inhibition of platelet P-selectin expression.
