目的:建立人体血浆中同时测定咪达唑仑和阿曲库铵HPLC测定法。方法:采用HPLC法,色谱柱为HypersilC18柱(250mm×4.6mm,5μm),流动相为甲醇-水(60∶40)(含0.1%二乙胺,0.05%磷酸溶液),流速为1.0mL·min-1,柱温为室温,检测波长225nm。结果:咪达唑仑在0.25~4μg·mL-1内呈良好的线性关系(r=0.9997),阿曲库铵在6.00~96.00μg·mL-1内呈良好的线性关系(r=0.9998),咪达唑仑和阿曲库铵最低检测浓度分别为50,1.5μg·mL-1,方法回收率在96.8%~100.0%,萃取回收率在87.7%~93.0%。日内、日间RSD(n=3)均小于5.9%。结论:本方法简便、准确,可靠,可用于人体血浆中同时测定咪达唑仑和阿曲库铵的浓度。
Objective: To establish a HPLC method for the simultaneous determination of midazolam and atracurium in human plasma. Methods: The HPLC column was Hypersil C18 column (250 mm × 4.6 mm, 5 μm). The mobile phase was methanol-water (60:40) containing 0.1% diethylamine and 0.05% phosphoric acid solution and the flow rate was 1.0 mL · min-1, the column temperature was room temperature, detection wavelength 225nm. RESULTS: Midazolam showed a good linearity (r = 0.9997) in the range of 0.25-4μg · mL-1 and a good linear relationship of atracurium in the range of 6.00-96.00μg · mL-1 (r = 0.9998) , Midazolam and atracurium were 50 and 1.5 μg · mL -1, respectively. The recoveries were 96.8% ~ 100.0%, and the recoveries were 87.7% ~ 93.0%. Day and day RSD (n = 3) were less than 5.9%. Conclusion: The method is simple, accurate and reliable and can be used for the simultaneous determination of midazolam and atracurium in human plasma.
