目的探讨Nanog在小鼠原始生殖细胞发育过程中的时空表达。方法采用免疫荧光技术定性研究Nanog在妊娠7.5~15.5d胚胎原始生殖细胞的表达;抗SSEA-1、抗Nanog双标记激光共焦扫描进一步定量分析妊娠11.5~15.5d小鼠原始生殖细胞中Nanog的表达变化。结果尿囊、后肠及背肠系膜、生殖嵴的原始生殖细胞中均可见Nanog的阳性表达。妊娠11.5d小鼠原始生殖细胞中Nanog的表达阳性率最高、荧光强度最强,雌性小鼠在妊娠12.5d后Nanog表达急剧下降,雄性小鼠在妊娠13.5d后Nanog表达急剧下降。结论Nanog在增殖的小鼠原始生殖细胞中稳定表达,其表达下调可能与原始生殖细胞的分化启动有关。
Objective To investigate the spatial and temporal expression of Nanog in mouse primordial germ cells. Methods Immunofluorescent technique was used to qualitatively study the expression of Nanog in primordial germ cells from 7.5 to 15.5 d of gestation. The anti-SSEA-1 and anti-Nanog double-labeled confocal laser scanning confocal microscope was used to quantitatively analyze the expression of Nanog in primordial germ cells from 11.5 to 15.5 d Change of expression Results The positive expression of Nanog was found in the primordial germ cells of the alveolar, hindgut and dorsal mesentery and genital ridge. The positive expression rate of Nanog was highest in the primordial germ cells of mice at 11.5 d after gestation, and the fluorescence intensity was the strongest. The expression of Nanog decreased sharply in female mice 12.5 d after gestation, and the expression of Nanog decreased sharply in 13.5 d after gestation. Conclusion Nanog is stably expressed in proliferating mouse primordial germ cells, and its down-regulation may be related to the initiation of differentiation of primordial germ cells.
