[目的]研究脐淋巴细胞与EBV转化淋巴细胞(LCIs),EBV相关细胞核基质蛋白(NMPs)之间的关系,分析鉴定EBV感染细胞后的NMPs特异性改变,探讨NMPs与EBV转化及肿瘤形成的关系。[方法]EBV转化淋巴细胞;改良方法提取NMPs;制备NMPs免疫小鼠血清;SDS-PAGE电泳及双波长蛋白薄层扫描方法分析NMPs;免疫蛋白印迹分析NMPs与抗NMPs的阳性血清之间的反应性。[结果]获得EBV转化淋巴细胞。获得五种细胞NMPs。各细胞间分子量相同的蛋白约占各自细胞组分的52.1%~59.6%。脐淋巴细胞30KD~40KD蛋白较多,而LCLs和EBV相关淋巴细胞中,高分子量的蛋白比较多。-64KD NMP特异地存在于LCLs及EBV相关细胞中。NPC病人血清中能够检出抗LCLs的NMPs抗体。[结论]LeuLeu-OMe和环孢菌素A结合使用,可促进EBV转化。低分化的脐淋巴细胞比高分化LCLs有更稳定的核基质网架结构。推测相同分子量NMPs构成核基质的结构成分;EBV感染细胞后,NMPs有明显的变化。NMPs的特异性与EBV感染高度相关;EBV转化与NPC的发生有一定关系。
[Objective] To study the relationship between umbilical lymphocytes and EBV transformed lymphocytes (LCIs) and EBV-associated nuclear matrix proteins (NMPs), analyze and identify the specific changes of NMPs after EBV infected cells, and explore the transformation of NMPs and EBV and tumor formation. relationship. [Results] EBV transformed lymphocytes were obtained. Five cell NMPs were obtained. The proteins with the same molecular weight among the cells accounted for about 52.1% to 59.6% of the respective cell components. Umbilical lymphocytes have more 30KD to 40KD proteins, while LCLs and EBV-associated lymphocytes have more high-molecular-weight proteins. -64KD NMP is specifically present in LCLs and EBV-associated cells. Anti-LCLs of NMPs antibodies can be detected in serum of NPC patients. [Conclusion] The combination of LeuLeu-OMe and cyclosporin A can promote the transformation of EBV. Poorly differentiated umbilical lymphocytes have a more stable nuclear matrix network structure than highly differentiated LCLs. It is speculated that the same molecular weight NMPs constitute the structural components of the nuclear matrix; after EBV infection of cells, NMPs have obvious changes. The specificity of NMPs is highly correlated with EBV infection; EBV transformation has a certain relationship with the occurrence of NPC.
