通过对不同分离株弓形虫抗原的分析 ,探讨弓形虫虫株间的差异性。取弓形虫 CAg阳性血液标本进行小鼠接种 ,对分离株进行生物学特性鉴定 ;采用流式细胞仪 ( FCM)对分离株弓形虫速殖子进行群体 DNA含量分析。结果在 5份 CAg阳性血液样本中分离到 4株弓形虫 ,两者符合率达 80 % ;经生物学鉴定结果显示 ,4个分离株的形态大小、毒力及抗原性均与 RH株弓形虫相似。上述 5株弓形虫速殖子 DNA含量分析均呈明显规律性 ,即均有 2个分布峰 ,但各虫株间的 2个分布峰中所占速殖子数存在显著性差异 ( P<0 .0 1)。表明检测弓形虫 CAg可作为弓形虫早期、急性期感染的确诊指标 ;采用流式细胞术进行 DNA群体分析 ,似可反映弓形虫虫株间的差异
Through the analysis of Toxoplasma gondii antigens of different isolates, the differences between Toxoplasma gondii strains were explored. Toxoplasma gondii CAg positive blood samples were used to inoculate mice and the biological characteristics of the isolates were identified. The DNA content of Toxoplasma gondii tachyzoites was analyzed by flow cytometry (FCM). Results Four Toxoplasma gondii isolated from 5 samples of CAg positive blood showed a coincidence rate of 80%. According to the biological identification results, the morphological, virulence and antigenicity of 4 isolates were similar to those of RH Toxoplasma gondii similar. The Toxoplasma tachyzoites DNA content analysis showed obvious regularity, that is, there are two distribution peaks, but the two distribution peaks among the tachyzoites there is a significant difference (P <0 .0 1). Toxoplasma gondii test showed that Toxoplasma gondii can be used as a diagnostic indicator of early and acute stage Toxoplasma gondii infection.
