目的:利用4种发根农杆菌R15834,R1601,R1000,A4诱导药用植物王不留行产生毛状根,建立王不留行的毛状根培养体系。方法:采用共培养法研究不同外植体、菌株、外源激素、侵染时间、预培养时间和共培养时间对王不留行毛状根诱导率的影响。结果:用王不留行叶片作为外植体,外植体预培养60 h,共培养48 h,发根农杆菌R1601侵染10 min,培养基中添加0.1 mg.L-1吲哚丁酸(IBA)转化率最高。结论:王不留行毛状根诱导成功,为其遗传转化提供了依据。
OBJECTIVE: To induce the hairy roots of medicinal plant king by using four kinds of Agrobacterium rhizogenes R15834, R1601, R1000 and A4 to establish the hairy root culture system of Wangbuyuhang. Methods: Co-culture method was used to study the effects of different explants, strains, exogenous hormones, infection time, pre-culture time and co-culture time on the induction rate of hairy roots. Results: The explants were pre-cultured with explants for 60 h, co-cultured for 48 h and Agrobacterium rhizogenes R1601 for 10 min. The medium was supplemented with 0.1 mg · L -1 indole butyric acid (IBA) the highest conversion rate. Conclusion: The success of hairy root induction by Wang Buyu provided the basis for its genetic transformation.
