目的评价用对照提取物定性和一测多评定量相结合的方法对白芷药材进行质量控制。方法用高效液相色谱法,色谱柱:ACE Ultra Core C_(18)(50 mm×2.1 mm,2.5μm),流动相:乙腈-水梯度洗脱,流速:0.5 mL·min~(-1),柱温:30℃,检测波长:310 nm,进样量:5μL。考察该方法的专属性、标准曲线和定量下限、精密度与回收率、稳定性以及重现性。结果高效液相色谱法对白芷中的5种主要香豆素类成分,佛手柑内酯、氧化前胡素、欧前胡素、cinidilin和异欧前胡素进行了快速分离,线性关系良好(r≥0.999 4),回收率较高,均在96.4%~100.0%内。对照提取物能快速对白芷中的5种主要成分进行定性。单标定量方法测得成分含量与标准曲线法定量的结果一致。结论对照提取物定性和单标定量相结合的质量控制方法是简单、可行的,能有效地对白芷药材进行质量控制。
OBJECTIVE To evaluate the quality control of Angelica dahurica by qualitatively combining the control extract and a multiple assessment quantity. Methods High performance liquid chromatography (HPLC) was used with a column gradient of ACE Ultra Core C 18 (50 mm × 2.1 mm, 2.5 μm). The mobile phase consisted of acetonitrile and water with a flow rate of 0.5 mL · min -1. , Column temperature: 30 ℃, detection wavelength: 310 nm, injection volume: 5μL. The method specificity, standard curve and quantitative limit, precision and recovery, stability and reproducibility were investigated. Results High performance liquid chromatography (HPLC) was used to separate the five major coumarins from Angelica dahuricae, bergapten, Oxidized imperatorin, Imperatorin, cinidilin and Isoimperatorin with good linearity r≥0.999 4), the recovery rate is higher, both within 96.4% ~ 100.0%. Control extract can quickly identify the five main components in Angelica qualitative. The content of the single-labeled quantitative method was consistent with that of the standard curve method. Conclusion The method of quality control combining qualitative and single-labeled quantitative analysis is simple and feasible, and can effectively control the quality of Angelica dahurica.
