水稻白叶枯病原细菌单克隆抗体杂交瘤细胞株的建立及初步应用

经水稻白叶枯病原细菌(Xanthomonas campestris pv.oryzae)菌株K—s—6—6免疫的BALB/C小鼠的脾细胞与骨髓瘤细胞SP2/0融合,经筛选和克隆化,获得3株稳定分泌抗该病原细菌单克隆抗体的杂交瘤细胞株k_1,K_2和K_(31)。分泌抗体稳定性试验、封闭试验、染色体数计数、交叉反应试验等表明具有株特异性;抗体分属IgG_3和IgG_(26)亚类;腹水抗体滴度在1:10~4左右。初步用于检测白叶枯病菌菌株,病叶和病种证明特异性高,匀质均一,在理论和生产上都有实用价值。 The spleen cells of BALB / C mice immunized with Xanthomonas campestris pv. Oryzae strain K-s-6-6 were fused with myeloma cells SP2 / 0 and screened and cloned to obtain 3 strains The hybridoma cell lines stably secreting monoclonal antibodies against the pathogenic bacteria k_1, k_2 and K_ (31). Secretion antibody stability test, blocking test, chromosome count, cross-reaction test showed that the strain-specific antibodies belong to IgG_3 and IgG_ (26) subclasses; ascites antibody titers in the 1:10 ~ 4 or so. Preliminary detection of bacterial blight bacterial strains, sick leaves and disease proved high specificity, homogeneous uniform, both in theory and production have practical value.
