以大豆未成熟子叶为外植体,采用基因枪轰击法进行遗传转化,探讨大豆基因型、受体轰击前预培养时间、轰击次数、轰击距离等因子对大豆未成熟子叶基因枪转化的影响。结果表明:大豆基因型对未成熟子叶基因枪转化有较大的影响,在参试的22个基因型中有40.91%的基因型体细胞胚胎发生率大于40%,胚数在2个以上,适宜用作大豆遗传转化的受体;合丰25未成熟子叶在轰击前预培养10 d优于预培养5和14 d。基因枪轰击1或2次对转化没有影响。基因枪轰击距离对转化的影响依大豆基因型而异。
The immature cotyledon of soybean was used as explant for genetic transformation by using gene gun bombardment method. The effects of soybean genotype, pre-incubation time before bombardment, bombardment times and bombardment distance on the gene gun transformation of immature cotyledon of soybean were investigated. The results showed that the soybean genotype had a great influence on the gene gun transformation of immature cotyledons. The incidence of somatic embryogenesis in 40.91% of the 22 tested genotypes was more than 40% and the number of embryos was more than 2, Which is suitable for the genetic transformation of soybean; Hefei 25 immature cotyledons before pre-bombardment 10 d better than pre-culture 5 and 14 d. One or two particle bombardment had no effect on transformation. The impact of the biolistic bombardment distance on transformation varies with the soybean genotype.
