目的:建立了一种简单、灵敏的酶催化动力学光度法测定盐酸多巴胺的新方法。方法:在pH9.8的NH3-NH4Cl的缓冲溶液中,利用盐酸多巴胺对牛血红蛋白(Hemoglobin,Hb)模拟酶催化体系的抑制作用,研究了该抑制反应的最佳实验条件及动力学行为。结果:测定的线性范围为1.1×10-7~1.1×10-5mol.L-1,检出限为6.1×10-8mol.L-1。对浓度为5.0×10-6mol.L-1的盐酸多巴胺进行11次平行测定,其相对标准偏差为4.5(。结论:该方法简单,快速,灵敏,可用于盐酸多巴胺注射液中盐酸多巴胺含量的测定。
OBJECTIVE: To establish a simple and sensitive new method for the determination of dopamine hydrochloride by enzymatic kinetic spectrophotometry. Methods: The optimum conditions and kinetics of the inhibition reaction were studied by using the inhibitory effect of dopamine hydrochloride on the hemoglobin (Hb) mimic enzyme catalyzed by NH3-NH4Cl in pH9.8. Results: The linear range was 1.1 × 10-7 ~ 1.1 × 10-5mol.L-1 with the detection limit of 6.1 × 10-8mol.L-1. The relative standard deviation (RSD) of eleven replicates of dopamine hydrochloride at a concentration of 5.0 × 10-6 mol·L-1 was 4.5. (Conclusion: The method is simple, rapid and sensitive and can be used for the determination of dopamine hydrochloride content in dopamine hydrochloride injection Determination.
