鞭毛蛋白是细菌鞭毛的主要成分,通过聚合鞭毛蛋白单体形成鞭毛的丝状体。鞭毛蛋白的N-末端和C-末端区域相互靠拢形成功能区域,该区域处于鞭毛丝状体的内部;中间区域是序列变异区,暴露于鞭毛丝状体表面。沙门菌鞭毛蛋白具有特殊的抗原性,称为H抗原,是沙门菌分型的依据之一。作为Toll样受体5(Toll-like receptors 5,TLR5)等先天性免疫系统受体的配体,沙门菌鞭毛蛋白是一种强效的蛋白型佐剂,已广泛应用于细菌、病毒和寄生虫的疫苗研制。本文就沙门菌鞭毛蛋白作为载体蛋白在偶联疫苗的应用作一综述。
Flagellin is a major component of bacterial flagella that forms flagellar filaments by polymerizing flagellin monomers. The N-terminal and C-terminal regions of flagellin are closed to each other to form a functional region that is inside the flagellar filaments; the middle region is a sequence-variant region that is exposed to the surface of the flagellar filaments. Salmonella flagellin has a special antigenicity, called H antigen, is one of the basis of Salmonella typing. Salmonella flagellin, a potent protein adjuvant, has been widely used as a ligand for innate immune system receptors such as Toll-like receptors 5 (TLR5) and has been widely used in bacteria, viruses and parasites Insect vaccine development. This review summarizes the application of Salmonella flagellin as a carrier protein in conjugate vaccines.
