目的 观察胃癌特异性转移因子 (GSTF)对淋巴细胞进行诱导刺激后 ,淋巴细胞表面的 IL- 2受体 (IL- 2 R)表达的变化 .方法 应用流式细胞仪检测异硫氰基荧光素 (FITC)标记的淋巴细胞表面 IL - 2 R抗体的荧光强度 ,以观察其受刺激前后表面 IL- 2 R的表达 .结果 GSTF在与其特异性抗原共同存在下诱导刺激淋巴细胞 ,可使其表面的 IL- 2 R表达量增加 ,且 GSTF的这种作用具有抗原特异性 ,GSTF或胃癌抗原单独存在时此作用则不明显 .结论 为肿瘤特异性转移因子的活性检测建立了一种新型快速、客观科学的检测方法 ,也为今后肿瘤特异性转移因子的分离纯化及进一步研究转移因子的作用机制打下了基础 .
Objective To observe the changes of IL-2 receptor (IL-2 R) expression on lymphocytes after lymphocyte stimulation with gastric cancer-specific transfer factor (GSTF). Methods Flow cytometry was used to detect isothiocyanate fluorescein (FITC) The fluorescence intensity of the IL-2 R antibody on the surface of the lymphocytes labeled to observe the expression of IL-2 R on the surface before and after stimulation. Results GSTF induces lymphocyte stimulation in the presence of its specific antigen together with its surface. The expression level of IL-2 R was increased, and this effect of GSTF was antigen-specific. This effect was not obvious when GSTF or gastric cancer antigen alone existed. Conclusion: A novel and rapid method has been established for the detection of tumor specific transfer factor activity. The objective and scientific detection methods also lay the foundation for the future separation and purification of tumor-specific transfer factors and the further study of the mechanism of transfer factors.
