研究了各种因素对柑桔种质材料的茎端、茎段外植体离体缓慢生长保存的影响。结果表明,培养物保存12个月时,在温度15℃或光照500Lx的条件下成活60%~70%,培养基成分中用2%甘露醇取代蔗糖的处理,成活率50%;暗培养和培养基成分中用高浓度甘露醇取代蔗糖不利于柑桔种质离体保存。对不同封口材料的比较研究表明,棉塞加聚乙烯膜可防止培养基水分减少,成活率较高,封口膜略次于前者。通过本研究建立了在一个缓慢生长条件下离体保存柑桔种质的方法。
The effects of various factors on the slow growth and preservation of stem explants and stem explants in citrus germplasm were studied. The results showed that when the culture was stored for 12 months, the survival rate was 50% when the culture was treated with 60% -70% of the culture medium under the conditions of 15 ℃ or 500Lx light irradiation and 2% mannitol instead of sucrose in the culture medium. Substitution of sucrose with high concentration of mannitol is not conducive to the preservation of citrus germplasm in vitro. Comparison of different sealing materials showed that the tampon plus polyethylene film can prevent the media moisture reduction, higher survival rate, sealing film slightly inferior to the former. Through this study, we established a method to conserve citrus germplasm in vitro under a slow growth condition.
