目的:构建肿瘤相关基因SNC90哺乳动物细胞表达型载体并进行转染研究.方法:肿瘤相关基因SNC90cDNA片断亚克隆至哺乳动物细胞表达型质粒pREP9中,并运用脂质体和电穿孔法将重组质粒pREP9-SN90转入3株人大肠癌细胞中,用G418筛选抗性细胞克隆.结果:重组体pREP9-SN90对SW1116,COLO205和SW620肠癌细胞株克隆形成均有一定的抑制作用,抑制率分别为72.2%,74.2%和59.7%.结论:这提示肿瘤相关基因SNC90对肠癌细胞生长起负性调节作用.
OBJECTIVE: To construct an expression vector for tumor-associated gene SNC90 mammalian cells and perform transfection. Methods: The tumor-associated gene SNC90 cDNA was subcloned into the mammalian cell expression plasmid pREP9, and the recombinant plasmid was prepared by liposome and electroporation. pREP9-SN90 was transferred into 3 human colorectal cancer cells and screened with G418. Results: The recombinant plasmid pREP9-SN90 had certain inhibitory effects on the colony formation of SW1116, COLO205 and SW620 colon cancer cell lines, and the inhibition rates were respectively. It was 72.2%, 74.2% and 59.7%. Conclusion: This suggests that tumor-associated gene SNC90 has a negative regulatory effect on the growth of colorectal cancer cells.
