本研究以棉花子叶为材料,探索一种适用棉花批量SSR-PCR分析的DNA快速提取方法。本方法只需要两次加液,两次高温水煮即可完成。紫外检测结果表明,所得DNA浓度较好,纯度较差,含有较多蛋白质。但SSR分析结果显示,可以扩增出相应的带型,且清晰易辨,准确可靠。相比常规CTAB法,具有操作简便,成本低,效率高,且无毒,无污染的优点,适用于大量棉花样品的SSR-PCR分析。
In this study, cotton cotyledons as material, to explore a cotton bulk SSR-PCR analysis of DNA rapid extraction method. This method requires only two times to add liquid, twice to complete high-temperature boiled water. The result of UV detection showed that the DNA concentration was better, the purity was lower and more protein was contained. However, the results of SSR analysis showed that the corresponding bands could be amplified and legible, accurate and reliable. Compared with the conventional CTAB method, it has the advantages of simple operation, low cost, high efficiency, no toxicity and no pollution and is suitable for SSR-PCR analysis of a large amount of cotton samples.
