目的:探讨胰蛋白酶抑制剂sporamin对人胰腺癌细胞增殖、迁移和侵袭及Notch4蛋白表达的影响。方法:采用MTT法检测sporamin对人胰腺癌细胞株PANC-1和MiaPaCa-2增殖能力的影响,Transwell法检测胰腺癌细胞体外迁移和侵袭能力,Western blotting检测不同质量浓度的sporamin(0、25、50、75、100μg/ml)对人胰腺癌细胞中Notch4蛋白表达水平的影响。结果:Sporamin能显著抑制人胰腺癌PANC-1和MiaPaCa-2细胞的增殖、迁移和侵袭能力,三种作用均呈显著质量浓度依赖性(P<0.05),但无显著时间依赖性(P>0.05);随着sporamin质量浓度的增加,Notch4蛋白水平的表达也逐渐下降。结论:胰蛋白酶抑制剂sporamin能质量浓度依赖性抑制人胰腺癌细胞增殖、迁移和侵袭能力,同时抑制人胰腺癌细胞中Notch4蛋白的表达。
AIM: To investigate the effect of sporamin, a trypsin inhibitor, on the proliferation, migration, invasion and the expression of Notch4 protein in human pancreatic cancer cells. Methods: The effects of sporamin on the proliferation of human pancreatic cancer cell lines PANC-1 and MiaPaCa-2 were detected by MTT assay. The migration and invasion ability of pancreatic cancer cells in vitro were detected by Transwell assay. The expressions of sporamin (0, 25, 50, 75, 100μg / ml) on the expression of Notch4 protein in human pancreatic cancer cells. RESULTS: Sporamin significantly inhibited the proliferation, migration and invasion of human pancreatic cancer PANC-1 and MiaPaCa-2 cells in a dose-dependent manner (P <0.05) 0.05). With the increase of sporamin concentration, the expression of Notch4 protein decreased gradually. CONCLUSION: The trypsin inhibitor sporamin can inhibit the proliferation, migration and invasion of human pancreatic cancer cells in a concentration-dependent manner and inhibit the expression of Notch4 protein in human pancreatic cancer cells in a concentration-dependent manner.
