1994~1996年对直接热处理苹果试管苗脱毒法进行了研究。将继代于MS+BA0.5~1mg1/L培养基上的苹果试管苗,置于光照培养箱内,每天照光14h,温度控制在37℃±1℃,热处理28天后,剥取0.5mm左右的茎尖进行培养,获得的试管苗用PAS-ELISA(A蛋白双夹心—酶联免疫吸附)法和指示植物法进行检测,其中有8个苹果品种脱去了苹果褪绿叶斑病毒(ACLSV)和苹果茎沟病毒(ASGV)。对ASGV和ACLSV的脱除率分别为81.8%和78.6%,说明直接对苹果试管苗进行热处理,再剥取茎尖进行培养是脱除苹果病毒,特别是ASGV行之有效的方法。
