本文研究低密度脂蛋白(LDL)对内皮细胞表达促炎性细胞因子白细胞介素6(IL-6)和肿瘤坏死因子(TNF)的影响及17β-雌二醇的保护作用。以体外培养的人脐静脉内皮细胞(HUVEC)为模型。将LDL与HUVEC孵育及HUVEC经它莫西芬(Tamoxifen)和/或17β-雌二醇预处理后,再与LDL孵育,检测培养液中IL-6和TNF活性。发现LDL可明显增加内皮细胞表达IL-6和TNF。LDL100μg/ml时,TNF为0.74±0.09(ng/ml),IL-6为9164±739(u/ml),与对照组相比P<001;经17β雌二醇预处理后,细胞在LDL作用下,IL-6和TNF活性与对照组相比无显著差异(P>005);它莫西芬和17β-雌二醇处理后,再加LDL,IL-6和TNF活性增高,与对照组相比有显著差异。实验结果表明:LDL可促进血管内皮细胞高表达促炎性细胞因子IL-6和TNF。雌激素可明显抑制LDL引起的内皮细胞高表达IL-6和TNF,从而产生对内皮细胞的保护作用。此作用可被它莫西芬拮抗。因此雌激素保护内皮细胞的作用,可能部分通过内皮细胞上雌激素受体。

This effect can be antagonized by tamoxifen. Therefore, the role of estrogen in the protection of endothelial cells may partially through the estrogen receptor on endothelial cells.
