目的:检测近交2系豚鼠肝癌细胞line10分泌的生长因子TuGF(s)对人肝上皮癌细胞(BEL-7402)生长的影响。方法:集落形成测定法检测TuGF(s)对BEL-7402细胞在软琼脂培养基内生长的影响。MTT法测定TuGF(s)对BEL-7402细胞在液体培养基内增殖的影响。结果:软琼脂培养基内培养时TuGF(s)对BEL-7402细胞无集落形成促进或抑制作用;单层培养的条件下,TuGF(s)对BEL-7402细胞在1×105个/ml接种细胞浓度下,具有促进细胞生长的作用;而在1×104个/ml细胞浓度下,则为抑制细胞生长的作用。结论:TuGF(s)对BEL-7402细胞体外增殖的影响依实验条件的不同具有复杂的生物学调节作用
OBJECTIVE: To investigate the effect of growth factor TuGF (s) secreted by line10 of inbred mouse hepatocellular carcinoma cell line 2 on the growth of human hepatic epithelial cancer cells (BEL-7402). METHODS: Colony formation assay was used to determine the effect of TuGF(s) on the growth of BEL-7402 cells in soft agar medium. The effect of TuGF(s) on proliferation of BEL-7402 cells in liquid medium was measured by MTT assay. Results: TuGF(s) did not promote colony formation or inhibition of BEL-7402 cells when cultured in soft agar medium; TuGF(s) inoculated BEL-7402 cells at 1×10 5 cells/ml under monolayer culture conditions. At the cell concentration, it has an effect of promoting cell growth; at a cell concentration of 1×10 4 cells/ml, it is an effect of inhibiting cell growth. Conclusion: The effect of TuGF(s) on the proliferation of BEL-7402 cells in vitro has a complex biological regulation depending on the experimental conditions.
