目的 探讨高温对肿瘤细胞周期的影响。方法 将肝癌腹水型瘤株HepA细胞分为 3 7℃、40℃、42℃和 44℃四个温度组。每一组分别在 1h、2h、4h和 8h四个时间点取材 ,经PI染色在流式细胞仪上检测四种温度条件下 ,四个时间段时HepA细胞在各细胞周期中的细胞数。结果 3 7℃时 ,细胞处于活跃的增殖状态 ,绝大部分细胞处于S及G2 /M期。 40℃时 ,细胞增殖更趋活跃 ,G1期细胞减少。 42℃时 ,细胞增殖受到明显抑制 ,G1期细胞增加 ,G2 /M期细胞减少。 44℃时 ,12h ,细胞增殖受到进一步的抑制 ,G1期细胞明显增加 ,G2 /M期细胞进一步减少 ;48h ,高温对细胞增殖的影响反而减弱 ,与 12h时相比 ,G1期细胞明显减少 ,G2 /M期的细胞有了明显的增加 ,甚至多于 42℃时。结论 温和性高温 (4 2 44℃ )对HepA细胞的增殖活动有明显的抑制作用 ,使HepA细胞主要滞留于G1期
Objective To investigate the effect of high temperature on tumor cell cycle. Methods HepA cells were divided into four temperature groups (37 ℃, 40 ℃, 42 ℃ and 44 ℃). Each group was drawn at four time points of 1h, 2h, 4h and 8h respectively. The number of HepA cells in each cell cycle was detected by flow cytometry at four different time points. Results At 37 ℃, the cells were in an active proliferative state, most of the cells were in S and G2 / M phases. At 40 ℃, cell proliferation more active, G1 phase cells decreased. At 42 ℃, cell proliferation was significantly inhibited, G1 phase cells increased, G2 / M phase cells decreased. At 44 ℃ for 12h, the cell proliferation was further inhibited, the cells in G1 phase increased significantly, and the cells in G2 / M phase decreased further. At 48h, the effect of high temperature on cell proliferation was weakened. Compared with 12h, G2 / M phase cells have a significant increase, even more than 42 ℃ when. Conclusions Mild hyperthermia (4 444 ℃) significantly inhibited the proliferation of HepA cells, leaving HepA cells mainly in the G1 phase
