目的:探究高压氧(HBO)通过调节缺氧诱导因子-1α(HIF-1α)对卵巢癌SKOV-3细胞恶性生物学行为的影响及其作用机制。方法:细胞培养完成后,首先将卵巢癌SKOV-3细胞分为对照组和HBO组,初步分析HBO对卵巢癌SKOV-3细胞生物学行为的影响以及对HIF-1α表达的影响。然后将余下的SKOV-3细胞按照随机数字表法分为3组:NC组、HBO组和HBO+HIF-1α组。HBO组在HBO条件下培养,HBO+HIF-1α组在HBO处理前通过转染过表达HIF-1α。采用qPCR、Western blotting、CCK-8、克隆形成、细胞划痕、Transwell实验检测HBO处理后HIF-1α对卵巢癌SKOV-3细胞恶性生物学行为的影响。结果:在HBO环境中培养24 h后,卵巢癌SKOV-3细胞的增殖、克隆形成、迁移和侵袭能力均受到明显抑制(n P<0.05),并且HIF-1α mRNA和蛋白水平也显著降低(n P<0.05)。HBO组SKOV-3细胞的增殖、克隆形成、迁移、侵袭和上皮-间充质转化(EMT)能力均显著低于NC组(n P<0.05);而HBO+HIF-1α组的上述指标均显著高于HBO组(n P<0.05)。过表达HIF-1α部分逆转HBO对SKOV-3细胞增殖、迁移、侵袭和EMT(n P<0.05)。n 结论:HBO可以在体外通过抑制HIF-1α的表达发挥对卵巢癌SKOV-3细胞的抑制作用。“,”Objective:To explore the effects and mechanism of hyperbaric oxygen (HBO) on the malignant biological behaviors of ovarian cancer cells by regulating hypoxia inducible factor-1α (HIF-1α).Methods:After cell culture, the SKOV-3 ovarian cancer cells were divided into control group and HBO group. The effects of HBO on the biological behaviors of SKOV-3 cells and the expression of HIF-1α were preliminarily analyzed. Then the rest of the SKOV-3 cells were divided into three groups: NC group, HBO group, and HBO+ HIF-1α group, according to random number table. The HBO group was cultured by using HBO; the cells in the HBO+ HIF-1α group overexpressed their HIF-1α by transfection before HBO treatment. The effects of HIF-1α on malignant biological behaviors of SKOV-3 cells after HBO treatment were detected by quantitative polymerase chain reaction (qPCR), Western blotting, cell counting kit-8 (CCK-8), clone formation, cell scratch assay, and Transwell migration assay.Results:After 24 h of culturing with HBO, the proliferation, cloning, migration, and invasion abilities of SKOV-3 cells were significantly inhibited (n P<0.05), and the levels of HIF-1α mRNA and protein also decreased significantly (n P<0.05). The cell proliferation, cloning, migration, invasion, and epithelial-mesenchymal transition (EMT) abilities of the HBO group were significantly lower than those of NC group (n P <0.05), while the above indicators of the HBO+ HIF-1α group were significantly higher than those of the HBO group ( n P<0.05). The overexpression of HIF-1α would partially reverse the effects of HBO on the proliferation, migration, invasion, and EMT of SKOV-3 cells.n Conclusion:HBO can exert an inhibitory effect on SKOV-3 ovarian cancer cells by inhibiting the expression of HIF-1α in vitro.
