利用微管吸吮技术 ,定量研究了肝癌细胞在IV型胶原 /层粘素复合裱衬表面上的粘附特性 ,在此基础上 ,利用两种细胞骨架干扰剂进一步探讨了细胞骨架系统对肿瘤细胞粘附力的影响。结果 :以正常肝细胞对照 ,用细胞松弛素D(0 .2 5~ 5μg/mL)处理后 ,两种细胞粘附力大大降低约 70 %~90 % ,而癌细胞降低幅度更大。用秋水仙素 (1~ 6 0 μg/mL)处理后 ,癌细胞的粘附力降低或趋于降低 ,而正常肝细胞则升高 14 0 %~ 50 0 % ,两种细胞呈不同的效应方式。结论 :细胞骨架系统与细胞的粘附性直接相关 ,癌细胞细胞骨架的异常可能是其粘附性异常的结构基础
Micropipette suction technique was used to quantitatively study the adhesion characteristics of hepatocellular carcinoma cells on the surface of type IV collagen/laminin composite lining surface. Based on this, two cytoskeletal interfering agents were used to further explore the cytoskeletal system on tumor cells. The effect of adhesion. RESULTS: After treatment with cytochalasin D (0.25-5 μg/mL) in normal hepatocyte control, the adhesion of both cells was greatly reduced by about 70% to 90%, while the cancer cells were more greatly reduced. After treatment with colchicine (1 to 60 μg/mL), the adhesion of cancer cells decreased or tended to decrease, while that of normal hepatocytes increased by 140% to 500%. Both cells showed different effects. the way. Conclusion : The cytoskeletal system is directly related to the adhesion of cells. Abnormality of the cytoskeleton of cancer cells may be the structural basis of its abnormal adhesion.
