目的:探讨人大肠癌细胞碱性成纤维细胞生长因子(bFGF)的表达、定位,并检查其受体。方法:采用免疫细胞化学染色、125I标记抗体的原位放射免疫分析及放射受体分析方法。结果:RPMI4788细胞产生的bFGF大部分蓄积于细胞核内,一部分存在于细胞膜上,并在细胞膜有其受体。结论:在肿瘤的发生发展过程中,肿瘤细胞产生的bFGF有可能起自泌性生长因子的作用,也可能直接参与核的功能。
Objective: To investigate the expression and localization of basic fibroblast growth factor (bFGF) in human colon cancer cells and examine its receptors. METHODS: Immunocytochemical staining, in situ radioimmunoassay of 125I-labeled antibodies and radioreceptor assays were used. RESULTS: Most of the bFGF produced by RPMI4788 cells accumulated in the nucleus, some of which were present on the cell membrane and had receptors on the cell membrane. Conclusion: During the development of tumors, bFGF produced by tumor cells may play a role in secretory growth factors and may directly participate in the nuclear function.
