目的:研究人参根在生长季的不同时期几种氧化还原酶的活力变化。方法:以五年生人参根为供试材料,采用中性磷酸缓冲溶液提取粗酶液。应用紫外分光光度法分别测定过氧化物酶(POD)、过氧化氢酶(CAT)、多酚氧化酶(PPO)和抗坏血酸过氧化物酶(APX)的活力。结果:POD和PPO活力在果后参根生长期之前维持在较低水平,随后快速上升,并于果后参根生长期达到整个生长季的最高值。CAT和APX分别在展叶期和果后参根生长期出现活力高峰。结论:人参根在展叶期和果后参根生长期活性氧代谢水平最高。
Objective: To study the changes of the activities of several redox enzymes in different periods of ginseng root during the growing season. Methods: Five-year-old ginseng root was used as test material, and the crude enzyme solution was extracted by neutral phosphate buffer solution. The activities of peroxidase (POD), catalase (CAT), polyphenol oxidase (PPO) and ascorbate peroxidase (APX) were determined by UV spectrophotometry. Results: The activities of POD and PPO remained low before the ginseng root growth stage and then rose rapidly, reaching the highest value throughout the growing season in the ginseng root growth stage. CAT and APX peaked vigorously at the leaf-setting and post-harvest root-ginseng growth stages respectively. Conclusion: The root of ginseng has the highest level of active oxygen metabolism at the leaf-setting and post-harvest ginseng roots.
