目的探讨茶多酚(TP)对人脐静脉内皮细胞(HUVEC)生长增殖的影响。方法采用二因素3×5析因设计及噻唑兰(MTT)比色法检测不同作用时间及剂量的TP对HUVEC的生长增殖作用,采用倒置相差显微镜观察HUVEC形态的变化。结果在24h时间段,120,160,240mg/L组,在48,72h时间段,80、120、160、240mg/L组与对照组吸光度有显著性差异。24,48,72h的半数抑制浓度分别是184.41,127.70,108.98mg/L。且TP作用时间与剂量之间有交互作用。结论TP能抑制HUVEC增殖且具有量效-时效依赖关系。
Objective To investigate the effects of tea polyphenols (TP) on the proliferation and proliferation of human umbilical vein endothelial cells (HUVECs). Methods Two-factor 3×5 factorial design and thiazolyl (MTT) colorimetric assay were used to detect the growth and proliferation of HUVEC with different time and dosage of TP. The morphology of HUVEC was observed by inverted phase contrast microscope. Results There was a significant difference in the absorbance between the 80, 120, 160, and 240 mg/L groups and the control group at the 24 and 48 hours, 120, 160, and 240 mg/L groups at 48 and 72 hours. The inhibitory concentrations at 24, 48, and 72h were 184.41, 127.70, and 108.98 mg/L, respectively. There is an interaction between TP action time and dose. Conclusion TP can inhibit HUVEC proliferation and has a dose-time-dependent relationship.
