研究人生长激素 (hGH)对大肠癌细胞增殖和分化的影响 .收集人大肠癌细胞株Lovo和LS 174 T(简称 174 ) ,以顺铂和 /或不同浓度的hGH体外培养细胞 ,2 4h后以流式细胞仪测定细胞增殖周期等指标 .结果显示hGH能诱导 174细胞S期数率非常显著地增高 (P <0 .0 1) ;Lovo细胞 10 0ng/mL组Go G1期参数下降 (P <0 .0 5) ,S期参数增加 (P <0 .0 1) ,2 0 0ng/mL组G2 M期百分数增高 .同时用顺铂和hGH处理后 ,两株细胞S期数率均显著下降或无明显上升 ,并显著抑制了Lovo细胞的增殖指数 (P <0 .0 5) ,提高了对 174细胞的DNA抑制率 (P <0 .0 1) ,并且导致 174细胞大量凋亡和死亡 .提示hGH在体外应用于人大肠癌细胞能够使处于DNA合成期的细胞增加 ,为提高肿瘤对周期时相特异性化疗药物的敏感性提供了基础 ,顺铂能够抑制hGH促DNA合成和细胞增殖的作用 ,并且导致细胞大量凋亡
To study the effect of human growth hormone (hGH) on the proliferation and differentiation of colorectal cancer cells. Collect human colorectal cancer cell line Lovo and LS 174 T (abbreviated as 174) and culture cells in vitro with cisplatin and/or different concentrations of hGH, 24 hours later. The flow cytometry was used to determine the cell proliferation cycle and other indicators. The results showed that hGH could induce a very significant increase in the S-phase rate of 174 cells (P < 0.01); the Go G1 phase parameters of the Lovo cell 100 ng/mL group decreased (P <0. 01). <0. 0 5), S phase parameters increased (P <0. 01), and the percentage of G2 M phase increased in the 200 ng/mL group. After treatment with cisplatin and hGH, the S phase of both cells was significant. The decrease or no significant increase significantly inhibited the proliferation index of Lovo cells (P < 0.05), increased the DNA inhibition rate of 174 cells (P <0.01), and led to the massive apoptosis of 174 cells and Death, suggesting that hGH can be applied to human colorectal cancer cells in vitro can increase the number of cells in the DNA synthesis phase.
