为寻找适用于分子标记快速检测作物种子DNA的方法。本研究采用改良CTAB法、SDS法和碱提法分别提取4种农作物(玉米、水稻、大豆和高粱)种子DNA,用于RAPD、SSR标记检测。结果表明CTAB法较SDS法更适合于提取玉米胚和高粱种子DNA用于RAPD检测;碱提法提取大豆和水稻种子DNA纯化后,用于RAPD检测得到清晰扩增条带。三种DNA提取方法对作物种子提取的DNA均适用于SSR标记检测。根据分子标记方法选择不同DNA的提取方法提取作物种子DNA,才能快速获得准确有效的分子标记检测结果。
To find a suitable method for rapid detection of crop seed DNA using molecular markers. In this study, the seeds of four kinds of crops (corn, rice, soybean and sorghum) were extracted by modified CTAB method, SDS method and alkaline extraction method, respectively, for detection of RAPD and SSR markers. The results showed that CTAB method was more suitable than SDS method for extracting DNA from maize embryos and sorghum seeds for RAPD detection. Alkaline extraction of DNA from soybean and rice seeds was used for RAPD detection to obtain clear amplified bands. The three DNA extraction methods are suitable for SSR marker detection of DNA extracted from crop seeds. According to the method of molecular marker selection, different DNA extraction methods can be used to extract crop seed DNA in order to obtain accurate and effective molecular marker detection results rapidly.
