目的 研究 9-顺维 A酸 (9- cis- retinoic acid,9- cis RA)诱导肺鳞癌细胞株 L 78凋亡作用及其与 Rb、Cy-clin D1 基因表达的关系 ,初步探讨其作用机制。 方法 体外培养肺鳞癌细胞株 L78,随机分为两组 ,实验组 :加入浓度为 5 μmol/ L 的 9- cis RA;对照组 :加入浓度为 0 .1%二甲亚砜。两组均培养 4 8小时后用免疫组织化学法检测 L78细胞中 Rb基因表达率 ,用流式细胞仪技术分别检测 Rb、Cyclin D1 基因表达率和肿瘤细胞凋亡率 ,并研究三者之间的相关关系。 结果 实验组肺鳞癌细胞株 L78细胞 Rb基因表达明显增强 ,Cyclin D1 基因表达降低 ,细胞凋亡率增高(P<0 .0 1)。 Rb基因表达率与细胞凋亡发生率呈正相关 (r=0 .85 4 ,P<0 .0 5 ) ,Cyclin D1 基因表达率与细胞凋亡发生率呈负相关 (r=- 0 .812 ,P<0 .0 5 )。 结论 9- cis RA可能通过 Rb基因表达增强和 Cyclin D1 基因表达降低途径 ,使细胞明显阻滞在 G0 / G1 期 ,并诱导肺鳞癌细胞株 L 78细胞凋亡。
Objective To investigate the apoptosis of human lung squamous cell carcinoma cell line L 78 induced by 9-cis-retinoic acid (9-cis-retinoic acid) and its relationship with Rb and Cy-clin D1 gene expression mechanism. Methods Lung squamous cell carcinoma cell line L78 was cultured in vitro and randomly divided into two groups. Experimental group: 9-cis RA with concentration of 5 μmol / L; control group: DMSO with concentration of 0.1%. Forty-eight hours after culture, the expression of Rb gene in L78 cells was detected by immunohistochemical method. The expression of Rb and Cyclin D1 gene and the apoptosis rate of tumor cells were detected by flow cytometry. The relationship between. Results The expression of Rb gene in L78 cells of experimental group was significantly increased, the expression of Cyclin D1 gene was decreased and the apoptosis rate was increased (P <0.01). Rb gene expression rate was positively correlated with the incidence of apoptosis (r = 0.854, P <0.05), Cyclin D1 gene expression rate was negatively correlated with the incidence of apoptosis (r = -0.812, P <0. 05). Conclusions 9-cis RA may induce apoptosis in L78 lung cancer cell line through enhancing the expression of Rb gene and decreasing the expression of Cyclin D1 gene in G0 / G1 phase.
